It's also possible they have a software firewall installed on the server itself. What i'm getting at is that I've seen this exact situation happen when a webserver has a software firewall that temporarily blocks IP's that it detects as malicious.
Given that it's your company website, there would be reason for multiple connections coming from your network to hit that server or cluster. What I would do is ask your web host to check both of your ISP's public IP's to see if they show up in the firewall logs. Even if they don't, it wouldn't hurt to have both of those white-listed.
If it can be reached outside of your network, it is something on your network preventing it and 9 out of 10 times it is your DNS causing the conflict. I had this same problem using the same domain internally for AD as our external website.
I had to add a cononical name for office to autuconfigure the exchange server info. What is your DNS setting? Also try to his sub page in your website.
Like for example. John - I had trouble with O on the last pc I setup in the office. The Autodiscover didn't work and I had a heck of a time. Ended up with a MS tech doing some manual setup. I wonder if this is all related? I also like the idea of seeing what happens when going to a sub page on the site. I'll try that next time it fails. Too bad, but at least I will no longer be nuts trying to find WTH is going on and why sites are not resolving. Hope that helps some of you with the same issue and thanks to user whose tip led me to the solution!

I tried dscacheutil -flushcache, but still no dice. Then this is not a DNS problem. Have you tried another browser?
